DeepCatch™ Broad-Range Nucleic Acid Concentration Kit

A specialized low-input nucleic acid concentration system for volume-limited downstream assays—not another routine cleanup kit.

The DeepCatch™ Broad-Range Nucleic Acid Concentration Kit is designed to rapidly concentrate purified DNA and RNA into ultra-low volumes while providing high recovery across a broad size range, including ultra-short fragments down to 10 nt. Developed for downstream assays where target nucleic acids are extremely dilute, this carrier-free system maximizes the final sample concentration without introducing exogenous nucleic acid interference.

PRODUCT OVERVIEW

Overcome the Input-Volume Bottleneck

Low-abundance cfDNA, cfRNA, and miRNAs are often recovered in volumes that exceed the input limits of NGS, ddPCR, qPCR, and RT-qPCR. As a result, only a portion of the available nucleic acid may enter each reaction.

Conventional concentration methods may lose ultra-short fragments, co-concentrate salts and inhibitors, or require larger elution volumes. The DeepCatch™ Broad-Range Nucleic Acid Concentration Kit concentrates up to 200 µL of purified DNA or RNA into as little as 5 µL while retaining fragments down to 10 nt and washing away residual soluble impurities. This enables more of the available nucleic acid to be used in volume-limited downstream assays.

Same Molecules. Smaller Volume. More Sample in Every Reaction.

Built for Concentration, not Routine Cleanup

Unlike precipitation, evaporation, or general-purpose cleanup workflows, DeepCatch™ is specifically designed to concentrate purified nucleic acids into ultra-low volumes while preserving broad fragment coverage. Its carrier-free capture and integrated wash steps help retain ultra-short species and reduce residual soluble impurities before downstream analysis.

KEY BENEFITS

Built for Low-Input, Volume-Limited Molecular Assays

  • Ultra-low-volume concentration Concentrate starting volumes of up to 200 µL into as little as 5 µL, enabling more of the available nucleic acid to enter each downstream reaction.
  • Broad-range retention down to 10 nt Retain short and ultra-short nucleic acids, including experimentally demonstrated recovery of 10 nt fragments.
  • RNA and DNA, single- and double-stranded Concentrate a broad range of nucleic acid types rather than being limited to RNA-only, DNA-only, or narrow oligonucleotide applications.
  • Carrier-free concentration No carrier RNA, carrier DNA, glycogen, LPA, or related additives are required, helping avoid exogenous nucleic acid background in sensitive downstream assays.
  • Preservation of mixed fragment populations Simultaneously retain ultra-short and longer nucleic acid fragments, supporting samples with complex size distributions.
  • Designed for limited-input downstream assays Increase the proportion of available nucleic acid introduced into NGS, ddPCR, qPCR, RT-qPCR, and other reactions with strict input-volume limits.
  • Wash-enabled concentration Magnetic bead wash steps help reduce residual soluble impurities rather than concentrating all buffer components together with the nucleic acids.
  • Improves Signal-to-Noise In representative applications, nucleic acid samples at pg/µL-level starting concentrations can be concentrated to ng/µL-levels while wash steps simultaneously clear away soluble inhibitors.

Performance Data

Verified Retention of Trace 10 nt fragments

Fig 1. Concentration of a trace 10 nt nucleic acid target

Fig 1. Concentration of a trace 10 nt nucleic acid target. A 10 ng FAM-labeled 10 nt oligonucleotide was diluted in 200 µL of low TE buffer and concentrated to a final volume of 5 µL using the DeepCatch™ Broad-Range Nucleic Acid Concentration Kit. Lane 1: 10 ng input; Lane 2: a 5 µL aliquot of the 200 µL sample before concentration; Lane 3: the entire 5 µL concentrated eluate. The concentrated eluate showed band intensity visually comparable to the input, demonstrating strong retention of the 10 nt oligonucleotide during a 40-fold reduction in sample volume. A faint, faster-migrating band in the input, consistent with free FAM dye, was not detected in the concentrated eluate, indicating that this low-molecular-weight non-nucleic-acid impurity was not co-concentrated under the tested conditions. Samples were analyzed by 15% denaturing urea-PAGE.

Broad-Range Retention Profile

Fig 2. Broad-Range Retention Profile

Fig 2. Ultra-short and longer nucleic acid fragments are retained together during concentration. A nucleic acid mixture containing 10, 15, 20, and 90 nt fragments was diluted in 200 µL of TE buffer and concentrated to 5 µL using the DeepCatch™ Broad-Range Nucleic Acid Concentration Kit. Lane 1: input mixture; Lane 2: a 5 µL aliquot of the 200 µL sample before concentration; Lane 3: the entire 5 µL concentrated eluate. Samples were analyzed by 15% denaturing urea-PAGE followed by SYBR Green staining. All tested fragment sizes, including the ultra-short 10 nt fragment, were readily detected in the concentrated eluate, demonstrating broad-range retention with no obvious size-dependent loss during a 40-fold reduction in sample volume.

Typical Applications

By drastically reducing the final sample volume, the kit enables a greater proportion of available nucleic acid to be added to reactions with strict input-volume limits. Recommended applications include:

  • Low-input NGS
  • Selected PCR-free NGS workflows
  • Liquid biopsy library preparation
  • Small RNA profiling
  • Rare-target ddPCR
  • High-sensitivity qPCR and RT-qPCR
